Salmonella Typhi

Salmonella Typhi is a human-specific pathogen causing the systemic febrile illness typhoid fever. We have completed the genome sequence of the well-studied strain Ty2, and compared it with that of the recently isolated Typhi strain CT18, sequenced at the Sanger Centre.

Whole-genome shotgun libraries of strain Ty2 were constructed in M13Janus (insert size ~2.0 kbp) and pBlueScript (insert size ~5 kbp). Clones were sequenced using dye terminator chemistry, collecting a total of 98,451 reads on ABI377 and 3700 instruments. A whole-genome optical map of NheI sites was also constructed. The completed genome (4,791,961 bp) has been annotated and deposited in the public databases (accession number AE014613).

Although many strains of Salmonella Typhi contain an IncHI1 plasmid, strain Ty2 contains no plasmids. We sequenced the prototype IncHI1 drug-resistance plasmid R27, originally isolated from Salmonella Typhimurium strain 1M1407. It is available from the public databases (accession number AF250878, erroneously reported as Salmonella Typhi).

Note: despite having different NCBI Taxonomy IDs, Salmonella typhi and Salmonella enterica subsp. enterica serovar Typhi are in fact the same species. This species is also referred to as Salmonella choleraesuis subsp. choleraesuis serotype Typhi and as Salmonella Typhi. See Salmonella nomenclature.

Strain availability

Salmonella Typhi strain Ty2 has been deposited with the American Type Culture Collection (ATCC) and is available as ATCC 700931, both as a culture and as purified DNA from this strain.

Sequence availability

We have placed the annotated genome sequence on our web server (see below). The chromosomal sequence has been processed by NCBI and entered into GenBank as 16 "pieces" (accession numbers AE016834 - AE016849), accessible via Entrez and BLAST. The complete genome as a single entry is also available via the NCBI ftp site.
